Abstract

Abstract Background and Objective: Salmonellosis is one of the most important food-borne bacterial zoonotic diseases worldwide, and poultry and its products are the major sources for salmonella transmission to human. Isolation of Salmonellaenterica from poultry needs bacteriologic enrichment and selected cultures of fecal samples. In this study, different culture methods for the isolation of salmonella from fecal samples were compared. Material and Methods: Forty- five positive samples from infected farms and 45 negative samples from normal farms were processed using enrichment media including tetrathionate broth, selenite cistine and Rappaport-Vassiliadis. Then the samples were incubated in selective cultures, and after 24 h, their results were compared with standard method. Results: Specificity of all methods for salmonella isolation was 100%, and salmonella was not isolated from the negative samples. The highest susceptibility was related to the method in which the sample first in Selenite cistine and later in Rappaport-Vassiliadis was enriched (100%). Enrichment in Rappaport-Vassiliadis could isolate 41 salmonella from 45 positive samples (91%) while the result of enrichment in tetrathionate was 6 isolates (13.3%). Conclusion: This study shows that enrichment in selenite cistine and then in Rappaport-Vassiliadis is currently the best method for isolating salmonella from fecal samples of poultry. Abstract

Abstract Background and Objective: Burkholderia cepacia complex (BCC) is a plant pathogen that is an important mortality factor in immune-compromised and hospitalized patients. We aimed to Isolate and Characterize the Burkholderia Cepacia Strains from Hospitalized Patients in the Hospitals of West Guilan Province. Material and Methods: This study was conducted on 90 saliva and blood samples obtained from patients with blood infection, pneumonia, asthma, patients connected to the monitoring and ventilator systems, and immune-compromised patients in different sections of hospitals such as the pediatrics, internal section, ICU and CCU. Primary screening was performed by cultivating the samples on Burkholderia cepacia selective agar (BCSA) Sensitivity to antibiotics was tested by Kirby-Bauer and Muller-Hinton Agar (MHA) and the separated isolations were recognized by strengthening the gene rec A. Results: Of 90 isolated samples, only one strain suspected B. cepacia was isolated from 24-year old women with asthma. This strain was resistant to bacitracin, pipracillin and ciprofloxacin antibiotics. Conclusion: The incidence of B. cepacia is rare in western part of Guilan province, which is congruent with the results of overall incidence in Iran.

Abstract

ABSTRACT
           Background and Objectives: Halophilic bacteria can grow and survive in environments with a wide range a wide range of salinities. In this study, we aimed to isolate halophilic bacteria from the Meyghan Lake in Arak (Iran) and evaluate their enzymatic activity.
           Methods: Samples were taken from four different areas of the lake. Halophilic bacteria were isolated by culture in moderate halophilic medium, sea water nutrient agar and nutrient agar containing different salt concentrations. Purification was done via consecutive culture methods, and production of extracellular hydrolytic enzymes including amylase, protease, lecithinase, DNase and lipase was evaluated.
           Results: Among 74 bacteria isolated from the lake water samples, 24 produced amylase, 27 produced lipase (Tween 40 and 80), 68 produced protease, three produced DNase and 61 produced lecithinase.
           Conclusion: In this study, we isolated halophilic bacteria with enzymatic activity and potential industrial applications.
